2nd Monolith was buried on the moon, requiring intelligence to discover it & be able to reach it. When it was dug out of the ground, it activated when the sun hit it, sending a signal to Saturn (Jupiter in the movie). Another test of intelligence.

In the book the 1st monolith was like a computer. It ran tests on the apes and interacted with them to find the smartest one to give knowledge of tools to. The bone was the first tool which gave those apes ascendency over it's enemies, starting evolution.
